    Smarter CARs: Using AI to Improve Chimeric Antigen Receptor Design

    Artificial intelligence is opening new possibilities in the design of next-generation immunotherapies. In this episode of EHA Unplugged, Dr Andrea Schmidts discusses how AI-driven approaches are reshaping the development of chimeric antigen receptor (CAR) T-cell therapies, particularly at the design stage of the CAR receptor binding domains.   Dr Schmidts explains how structure-based AI models can generate entirely new protein binders, moving beyond the traditional reliance on monoclonal antibody–derived fragments. The conversation explores how these tools enable large-scale exploration of CAR designs, the experimental challenges of validating thousands of candidates, and the potential to tailor therapies that overcome resistance mechanisms such as antigen escape.     What is Real-World Evidence Revealing About CAR T Therapy?

    In this episode, Prof Benoit Tessoulin examines how real-world evidence from the French DESCAR-T registry is expanding our understanding of CAR T-cell therapy beyond clinical trials. He explains how the registry captures data from patients considered for CAR T-cell treatment, including those who do not ultimately receive therapy, and explores what these data reveal about patient selection, treatment outcomes, bridging therapy, and comparisons between different CAR T-cell products.   The discussion also examines how real-world populations differ from clinical trial cohorts, in what measure registry data have confirmed the effectiveness of CAR T-cell therapy in routine practice, and what has been learned about toxicities such as CRS, ICANS, and infections. Prof Tessoulin highlights the importance of timely referral, evolving product selection strategies, and the growing role of patient registries in informing clinical decision-making and future research.     How to Start a Clinical Trial?

    What does it take to design and lead a clinical trial in hematology? In this episode of EHA Unplugged, Prof. Elias K.     How Experts Treat Multiple Myeloma

    In this episode, Prof. Pieter Sonneveld is joined by Prof. Meletios Dimopoulos and Prof. Evangelos Terpos to discuss the updated EHA–EMN (European Myeloma Network) guidelines for the diagnosis, treatment and follow-up of multiple myeloma.  The conversation explores how recent therapeutic advances—including 14 newly approved treatment regimens, CAR T-cell therapies, bispecific antibodies and antibody-drug conjugates—have reshaped the treatment landscape and informed the latest recommendations.     Menin Inhibitors to Treat Acute Myeloid Leukemia (AML)

    Get up to date on menin inhibitors, an emerging class of targeted therapies for acute myeloid leukemia (AML). Menin is a key component of the KMT2A (lysine methyltransferase 2A, formerly MLL1) complex, which plays a critical role in gene regulation, development, and cell cycle control. Small-molecule menin inhibitors have shown particular promise in KMT2A-rearranged (KMT2Ar) and NPM1-mutated AML, with two agents already receiving FDA breakthrough therapy designation.  In this episode, Dr Scott Armstrong and Prof Marc Raaijmakers explore the scientific rationale behind menin inhibition, identify which AML patient populations are most likely to benefit, and review the current clinical evidence supporting their use in defined molecular subtypes.     How Experts Treat HIV-associated Lymphomas

    Get up to date with the EHA–ESMO guidelines on Human immunodeficiency virus (HIV)-associated Lymphomas.  In this episode, Prof. Kai Hubel, a key author of the guidelines, explains the importance of having dedicated recommendations for patients with HIV-associated lymphomas.
