Can Patients Choose the Next PKD Research Targets? YES. Dr. Benjamin Freedman Explains

In this episode, Felix welcomes Dr. Benjamin “Beno” Freedman, Director of the PKD Invent Center at the University of Washington and one of the leading researchers in the world of PKD organoids — tiny “mini-kidney” structures grown from stem cells that can form PKD-like cysts in a dish.Beno explains how kidney organoids are made, why they are different from simple cell cultures or animal models, and how they allow researchers to study human PKD biology in a way that was not possible before. Felix and Beno go deep into the strange and fascinating details: cysts that grow “inside out,” extracellular matrix changing cyst orientation, myosin acting like a contractile force that may help cysts keep their shape, and the possibility that PKD cells may be mechanically weaker than normal cells. They also discuss one of the most relevant findings for the Reversing PKD community: how PKD organoids can pull in glucose through SGLT transporters, creating an osmotic pull that brings water into cysts. This connects directly to the larger metabolic picture of PKD, glucose exposure, SGLT2 inhibitors, ketogenic nutrition, and why lowering the glucose pressure on PKD tissue may matter. The second half of the conversation focuses on the new PKD Research Resource Consortium center and the possibility of patient-centered research input. Beno explains how scientists, companies, and potentially even the wider PKD community may be able to suggest molecules, supplements, etc. for pilot testing in PKD organoid systems.Viewers will learn:• What kidney organoids are and why they matter for PKD• How scientists grow “mini kidneys” from stem cells• Why PKD cysts can form differently depending on their environment• How extracellular matrix may change cyst orientation• Why myosin and cell contractility could become a therapeutic target• How glucose transport may help PKD cysts pull in water and expand• What SGLT transporters have to do with PKD research• How the new PKD Invent Center and PKD-RRC may allow community-driven research ideas• Why patient input could help shape the future of PKD research🎙️ Guest & Research Links• PKD Research Resource Consortiumhttps://pkd-rrc.org/• PKD Invent Center / University of Washingtonhttps://pkd-rrc.org/centers/invent/• Dr. Benjamin Freedman / Freedman Labhttps://freedmanlab.org/• Plurexa — Organoid-based drug discovery and research serviceshttps://www.plurexa.com/📚 Topics Mentioned• Kidney organoids• PKD organoids• Polycystic Kidney Disease in a dish• Stem cells and organogenesis• Extracellular matrix• Inside-out cyst formation• Myosin and cell contractility• SGLT transporters• Glucose-driven cyst expansion• Osmosis and cyst fluid accumulation• SGLT2 inhibitors• Patient-centered PKD research• Community screening of compounds and molecules🧬 This is not medical advice.🎧 Don’t forget to like, comment, and subscribe if this episode brought value to your PKD journey.Go tohttps://www.reversingpkd.comto find more learning resources and get on the PKD health coaching program.Find my book Reversing Polycystic Kidney Disease here:https://reversingpkd.com/booklinks

Cancer’s Hidden Trigger: How Fungi Ignite ALL Tumors & What It Could Mean for PKD - Mark Lintern

In this episode, Felix welcomes Author and citizen scientist Mark Lintern—the man who might finally have nailed what actually flips healthy cells into the Warburg Effect metabolism that Professor Thomas Seyfried made famous— and explained on the Reversing PKD Podcast. Mark lays out his “Cell Suppression Theory,” showing how fungi seize control of cells, and create the glucose-dependent breakdown of respiration that Seyfried’s metabolic theory describes by keeping cells stuck in the „Cell Danger Response“ (CDR). Not in error, but it an attempt to rescue the cell. The two swap notes on the parallels between malignant tumors and expanding kidney cysts: a terrain drenched in toxic triggers and suppressed E-cadherin, mirroring the endotoxin fingerprints Felix had already traced in Polycystic Kidney Disease prior. They explore how viewing both diseases as slow-motion reactions to infections and/or toxins reframes everything—from a ketogenic “first line of defense" to antifungals and antiparasitic tactics that could stop tumors and cysts at their microbial root.🎙️ Guest & Core Material• Mark Lintern – Cell Suppression Theory websitehttps://www.cellsuppression.com/ • Free 13-page synopsis (PDF)https://www.cellsuppression.com/marks-theory-synopsis-free-download • Book — The Cancer Resolution ? Beyond Glucose: How Glutamine Drives PKD and Cancer Growth

In this special episode, Felix sits down with renowned researcher Professor Thomas Seyfried to discuss a groundbreaking revelation for Polycystic Kidney Disease (PKD) management: it's not just glucose that fuels PKD cell growth—glutamine is a major player, too. While many PKD patients have now started to focus on ketogenic diets to restrict glucose, this interview uncovers why glutamine might be just as crucial to take into account. Professor Seyfried explains how glutamine drives cancer cell fermentation and uncontrolled growth, drawing parallels to PKD research. Together, Felix and Professor Seyfried explore how combining a ketogenic diet with glutamine targeting could open new doors for slowing PKD progression. Key topics covered: Why glutamine is a second fuel source for PKD cells. The role of mitochondrial dysfunction in both PKD and cancer. Insights from Professor Seyfried’s decades of research into metabolic therapies. If you're already using a ketogenic diet for PKD, this interview offers a new perspective on your results. Learn from one of the leading experts in metabolic disease research and take your knowledge about PKD to the next level.
