    Association of nonfribrillar amyloid oligomers with drusen of aging and age-related macular degeneration

    Protein misfolding and aggregation are thought to underlie the pathogenesis of many „amyloid diseases“, such as Alzheimer and Parkinson diseases, whereby a stepwise protein misfolding process begins with the conversion of soluble protein monomers to prefibrillar oligomers and progresses to the formation of insoluble amyloid fibrils. Drusen are extracellular deposits found in eyes afflicted with age-related macular degeneration (AMD). Recent characterizations of drusen have revealed protein components that are shared with amyloid deposits. However, characteristic amyloid fibrils have thus far not been identified in drusen. In this study, we tested the hypothesis that nonfibrillar oligomers may be a common link in amyloid diseases. Oligomers consisting of distinct amyloidogenic proteins and peptides can be detected by a recently developed antibody that is thought to recognize a common structure. Notably, oligomers exhibit cellular toxicity, which suggests that they play a role in the pathogenesis of neurodegenerative diseases. Through use of the anti-oligomer antibody, we came to observe the presence of nonfibrillar, toxic oligomers in drusen. Conversely, no reactivity was observed in age-matched control eyes without drusen. These results suggest that amyloid oligomers may be involved in drusen biogenesis and that similar protein misfolding processes may occur in AMD and amyloid diseases.

    Bakteriämie bei Hunden und Katzen mit Verdacht auf Sepsis - eine retrospektive Untersuchung

    In der vorliegenden Arbeit wurden Daten von 938 Hunden und 292 Katzen mit Verdacht auf Sepsis erfasst, bei denen im Zeitraum von 1995 bis 2004 an der Medizinischen Kleintierklinik der Ludwig-Maximilians-Universität in München Blutkulturen durchgeführt wurden. Ziel des ersten Teils der Studie war es, die Bakterienverteilung bei Hunden und Katzen mit Sepsisverdacht zu bestimmen und deren Antibiotikawirksamkeit zu ermitteln. Im zweiten Teil der Studie wurden die Daten der Hunde und Katzen mit Anzeichen einer Sepsis ausgewertet, bei denen ein klinisch signifikantes Bakterium aus dem Blut isoliert wurde oder bei denen ein möglicher Kontaminant aus mindestens zwei bakteriologischen Blutkulturen kultiviert wurde. Die klinischen und labordiagnostischen Veränderungen, die betroffenen Organsysteme, der Primärherd der Infektion und die Letalitätsrate dieser Hunde und Katzen wurden bestimmt. Von allen untersuchten Blutkulturen waren 24.4 % der Proben von Hunden und 22.6 % von Katzen positiv. Mischinfektionen mit verschiedenen Bakterien wurden bei 11.4 % der Hunde und 12.1 % der Katzen gefunden. Sowohl bei Hunden als auch bei Katzen überwogen grampositive mit 68.2 % bei Hunden und 50.7 % bei Katzen über gramnegative Bakterien. Die prozentual am häufigsten grampositiven Bakterien stellten Staphylococcus spp. und Streptococcus spp. dar, während die häufigsten gramnegativen Bakterien Escherichia coli waren. Enrofloxacin, Chloramphenicol, Cephalexin und Amoxicillin/Clavulansäure zeigten die höchste in-vitro-Gesamtwirksamkeit bei Hunden und Katzen. Unter den Antibiotikakombinationen zeigte die Kombination von Enrofloxacin mit Amoxicillin/Clavulansäure die beste Wirksamkeit. Befunde von 140 Hunden und 39 Katzen mit Sepsis wurden ausgewertet. Gastrointestinale Symptome und Anorexie waren die häufigsten vorberichtlichen Beschwerden bei Hunden und Katzen. Die häufigsten Laborveränderungen bei Hunden waren Neutrophilie, Linksverschiebung, Monozytose und Erhöhung der Alkalischen Phosphatase; wohingegen bei Katzen Neutrophilie, Linksverschiebung, Lymphopenie und Hyperglykämie am häufigsten zu finden waren. Bei 81.7 % der Hunde und 59.5 % der Katzen wurden mindestens zwei der „systemic-inflammatory-response-syndrome“-Kriterien gefunden. Disseminierte intravasale Koagulopathie trat bei 16.7 % der Hunde und bei 14.3 % der Katzen auf. Statistisch signifikant mehr Hunde mit gramnegativer Bakteriämie als mit grampositiver wiesen eine Hypalbuminämie auf (p = 0.007). Es gab keine anderen signifikanten Unterschiede zwischen Tieren mit gramnegativer und grampositiver Bakteriämie. Bei 30.7 % der Hunde und 23.1 % der Katzen konnte ein Primärherd der Infektion gefunden werden. Sowohl bei Hunden als auch bei Katzen wurden als häufigste Primärherde Hautinfektionen und der Gastrointestinaltrakt gefunden. Die Letalitätsrate betrug bei Hunden 32.6 %, bei Katzen 54.1 %.

    Die Behandlung metabolischer Azidosen bei Kälbern durch orale Gabe von Natriumbikarbonat unter Berücksichtigung des Pansensaft-pH-Wertes

    Das Ziel der vorliegenden Arbeit war es, herauszufinden in wieweit die Therapie einer metabolischen Azidose bei Durchfallkälbern durch die orale Verabreichung von Natriumbikarbonat per Kälberschlundsonde möglich ist und ob der Pansensaft-pH-Wert Einfluss auf die Resorption des in den Pansen eingegebenen Puffers hat. Es sollte festgestellt werden, ob es eine bestimmte für alle Kälber mit einer gering- bis mittelgradigen metabolischen Azidose geltende Menge an Natriumbikarbonat gibt, die oral verabreicht in der Lage ist diese Durchfallkälber zu therapieren, ohne dass Nebenwirkungen des Natriumbikarbonats auftreten. Das Patientengut bestand aus 32 bis zu vier Wochen alten Kälbern, die infolge eines Durchfallgeschehens eine metabolische Azidose aufwiesen. 50 g Natriumbikarbonat oral verabreicht war eine Puffermenge, die den Blut-pH-Wert und den BE-Wert der Kälber zumindest vorübergehend ausgeglichen hat, ohne dass deutliche Nebenwirkungen auftraten. Bei einigen Tieren dieser Untersuchung mussten weitere Pufferbehandlungen durchgeführt werden. Es gibt aber keinen eindeutigen Befund, der bereits im Voraus die Vermutung zulässt, welche Tiere eine weitere Pufferbehandlung benötigen. Bei Kälbern, die nach der Behandlung eine gute Tränkeaufnahme zeigen, ist eine ausreichende Wirkung der einmaligen Pufferbehandlung anzunehmen. Ein Einfluss des Pansensaft-pH-Wertes auf die Resorption des Natriumbikarbonats im Pansen konnte nicht festgestellt werden.

    Die Immunantwort boviner und capriner Euterepithelzellen auf verschiedene Mastitiserreger

    Infections of the bovine mammary gland are, in addition to the affliction of the animals, a great economic burden in the dairy industry. A better understanding of the innate immune response of the host could lead to a better selection of mastitis resistant cows or to better prophylactic and therapeutical treatments. The objective of the present study was to investigate the involvement of the epithelial cells into the outcome of mastitis induced by different pathogens. Primary epithelial cell cultures isolated from milk of Brown swiss cows and coloured german goats, and tissue of cows were used to test the immune response. Because the cells of the cows showed different responses to isolated bacterial endotoxins (lipopolysaccharide, lipoteichoic acid, and peptidoglycans) compared to whole bacteria, they were treated with heat inactivated (10 MOI) gram-negative Escherichia coli (E.coli), a very common pathogen causing acute and severe intra-mammary infections, with Staphylococcus aureus (S.aureus) and coagulase-negative Staphylococcus (CNS), both prevalent cause of chronic and subclinical, and, Streptococcus uberis (Str.uberis) an inducer of acute and chronic mastitis. Goat cells were treated with 10 MOI heat inactivated S.aureus causing in opposite to cows an acute, clinical mastitis and LPS. In cows E.coli induced an increased mRNA expression of interleukin-(IL) 8, IL-6, tumor necrosis factor alpha (TNF) and granulozytes, macrophage-stimulating factor (GM-CSF) within a 30 min treatment. IL-8, IL-6 and TNF mRNA levels were still elevated after 60 min, 90 min, 6 h and 24 h. After 6 h S.aureus induced an increase in mRNA expression of IL-6, IL-8, TNF and SAA. After a 24 h treatment the expression of these immunomodulators was still elevated. Interestingly, Str.uberis in the same concentration did only induce the mRNA expression of GM-CSF after 60 min and the IL-8 after a 6 h treatment but had no influence on other immunomodulator mRNA expression. CNS treated cells needed a MOI of 50 for any reaction. This MOI induced a reaction on mRNA levels of IL-6 and IL-8 but not on TNF after 6 h. In goats S.aureus induced an increase in mRNA expression of IL-8 and TNF after 1 and 6 h. After a 24 h treatment just the mRNA level of TNF was elevated. In conclusion mammary gland epithelial cells are involved in the different immune response to various mastitis pathogens. In cows, E.coli always induced an earlier immune response compared to S.aureus. This pathogen induced a later and probably for the immne system weaker reaction, not strong enough to cause an acute intramammary infection. Str.uberis in the same concentration induced only an increased GM-CSF and IL-8 mRNA expression after a 60 min or a 6 h treatment, respectively. This bacterium probably stimulates different immune relevant gene expressions not measured here or does not stimulate the mammary gland epithelial cells. The cells needed a 5-fold higher dose of KNS to respond with induced mRNA levels of IL-6 and TNF but not IL-8. KNS maybe does not stimulate the immune system enough to induce an acute mastitis and is like by the epithelial cells just recognized in very high dosis. IL-8 plays an important role as a chemotactic stimulus that is missing here. Goat mammary gland epithelial cells showed a early increase of IL-8 and TNF, that could be high enough, combined with the response of the immune cells in the milk, to induce an acute and severe intra-mammary infection.

    Entwicklung und Charakterisierung von Enzymimmuntests für den Nachweis von Fluorchinolonen

    Die vorliegende Arbeit befasst sich mit der Entwicklung enzymimmunologischer Verfahren zum Nachweis von in der EU-weit geltenden Verordnung (EWG) Nr. 2377/90 reglementierten Chinolonen. Zur Gewinnung gruppenspezifischer Antikörper wurde Ciprofloxacin-Ethylendiamin an Keyhole Limpet Haemocyanin (KLH) gekoppelt und drei Kaninchen intracutan immunisiert. Nach einer Restimulierung waren bei allen drei Kaninchen spezifische Antikörper nachweisbar. Die zur Erstellung eines direkten und indirekten kompetitiven enzymimmunologischen Nachweisverfahrens notwendigen markierten Antigene wurden durch Kopplung von Ciprofloxacin, Ciprofloxacin-Ethylendiamin, Clinafloxacin sowie Norfloxacin an Meerrettichperoxidase, Glucoseoxidase oder Ovalbumin hergestellt. Dabei kamen verschiedene Kopplungsmethoden (aktive Estermethode, Perjodatmethode und Carbodiimidmethode) zum Einsatz. Mit dem spezifischen Serum eines Tieres wurden verschiedene kompetitive Enzymimmun-testsysteme entwickelt. Von den direkten Enzymimmuntests wies das empfindlichste System eine Nachweisgrenze für Ciprofloxacin von 1,25 ng/ml auf, im indirekten Testsystem lag die untere Nachweisgrenze bei 1,5 ng/ml. Ciprofloxacin konnte somit weit unterhalb des geltenden MRL-Wertes von 100 µg/kg in Milch nachgewiesen werden. Das Antiserum zeigte nur bedingt Gruppenspezifität. Für fünf von 23 überprüften Chinolonen wurden Kreuzreaktionen von > 50 % ermittelt, von den mit einem MRL-Wert belegten Substanzen wurden Danofloxacin, Ciprofloxacin, Enrofloxacin und Oxolinsäure erfasst. Zur Überprüfung der Anwendbarkeit der entwickelten Testsysteme wurden künstlich kontaminierte und gewachsene Milch- und Garnelenproben untersucht, wobei das jeweilige Testsystem an die entsprechende Probenmatrix adaptiert wurde. Die mittlere Wiederfindungsrate lag bei 95,5 %, in keiner der Praxisproben konnten Chinolonrückstände nachgewiesen werden. Weiterhin wurde ein mikrobiologisches Verfahren unter Verwendung von E. coli als Testkeim zum Nachweis für Chinolone entwickelt. Dieser Agardiffusionstest zeichnete sich durch eine hohe Sensitivität und Gruppenspezifität aus. Ciprofloxacin konnte mit einer Nachweisgrenze von 2 ng/ml weit unterhalb des geltenden MRLs nachgewiesen werden, zehn weitere wichtige Chinolone wurden unterhalb von 100 ng/ml erfasst. Dieses System kann somit als nützlicher Kontrolltest verwendet werden.

    Epidemiologische und laborexperimentelle Untersuchungen zur Urolithiasis bei Katzen

    A long-term study (1981 – 2000) of the epidemic situation of urolithiasis in cats in Europe is presented in the first part of the work. Altogether 1.797 urinary stone analyses were established with the help of infrared spectroscopy of cats. It was struvite that was most often registered (64,2 %), followed by calcium oxalate (26 %). Proteine stones (1 %) were proved only within our investigation. The following breeds were mainly presented: European Shorthair (i. e. domestic shorthair) on the basis of their share in the whole population and Persian because of their predisposition to urolithi-asis. Calcium oxalate incidence increased from 2,2 – 5 % (1981 – 1990) to 33 % during the period of 1991 – 2000. Especially Persian, Chartreux, British Blue, Birman, Exotic Shorthair, Himalayan, Burmese, Somali und Ragdoll are susceptible to calcium oxalate formation. The average age of all involved cats was about 6,85 concerning female cats and 6,47 concerning male cats, the weight ─ about 5 kg for female cats and 5,4 kg ─ for male cats. More than 50 % of pets (according to the information given by the treating veterinary surgeons) were obese. During the investigation 55,1 % male and 44,9 % female cats were registered. The amount of castrated cats made up 88,8 %. The average rate of relapse of all kinds of urinary stones was about 27,6 %. The urinary calculi were located mainly in the bladder and/or in the urethra. The surgical ablation took place in 82,8 % of cases. 77,9 % of urinary stones were accompanied by lower urinary tract inflammation. In 91 % of cases it was the matter of infection. Most of the examined cats were fed with dry- (30,1 %) or canned- and dry food. It is importatant to underline that 42 % of all calcium oxalate stone patients were given only dry food. The urinary stones appeared in 40 % either as pure urinary semolina or as the combination of semolina and urinary stones. In the second part of this work 142 spontaneous urine samples of healthy cats were quantitatively analysed and statistically evaluated. Thus the referential area for the following urinary parameters of cats (relevant to calculus formation) was made: calcium, oxalic acid, magnesium, citric acid, phosphate, natrium, kalium, ammonium, sulphate, chlorine, uric acid, creatinine, urea, specific gravity, urinary pH and glyco-saminoglycan (GAG). And besides the relative supersaturation with calcium oxalate (RSCaOx = 1,63) and struvite (RSStru = 0,451) were assessed on the basis of urinary analyses with the help of EQUIL 2. Synthetic urine was produced on the basis of the established data (in the origi-nal research) of the composition of native cat’s urine. It was used for the crystallization tests of the formation of urinary stones in cats. The synthetic urine for cats consisted of the following components: calcium, oxalic acid, magnesium, citric acid, phosphate, natrium, kalium, ammonium, sulphate, chlorine, uric acid, creatinine and urea. The artificial urine did not possess any tendency to an independent change and was well reproductive. It was checked with the help of the synthetic cat’s urine in 3 test models the influence of the most important for stone formation substances (calcium, oxalate und magnesium) within their physiological arias in connection with urinary pH on the urolith formation. The results were presented in diagrams. The increase of the concentration of calcium in the carried out tests resulted in no calcium oxalate crystallization. However, just a small rise of oxalate (from [Ox] = 1,8 mmol/l) in acidic milieu (from pH = 5,5) caused the formation of calcium oxalate. The rise of magnesium concentration (over [Mg] = 3 mmol/l) prevented the formation of calcium oxalate. On average the crystallization of calcium oxalate in “oxalate-” and “magnesium model” began at RSCaOx = 6,65 ± 0,44 (EQUIL 2). The maximum RSCaOx, the synthetic cat’s urine could achieve without becoming solid, was 6,0 ± 0,32. Regardless of the concentration of the substances that were checked, struvite fell out at urinary pH 7,5 in all model series. The increase of the concentration of calcium (from [Ca] = 0,9 mmol/l) and urinary pH resulted in the crystallization of different phosphate phases (brushite and carbonate apatite). The results of the epidemiological and experimental tests are the basic know-ledge for the further work. With their help it is possible to test the influence of different inhibitors and promoters on the crystallization of calcium oxalate.

    Expression von lokalen Regulationsfaktoren im Ovar des Schweins

    The aim of the study was to further investigate the importance of local other and to detect possible differences of expression and localisation in poly- and monoovulatory species. Therefore ovaries of sexually mature gilts were collected at the local abattoir and follicles, which were determined to be in the follicular phase of oestrous cycle, ovarian stromal tissue and corpus luteum (CL) were prepared. In two experiments mRNA expression of gonadotropin receptors (LHR, FSHRregulatory factors during the final follicle growth in pig, to clarify relations among each), Aromatase (P450 Aro) and growth factors (fibroblast growth factor (FGF-1, FGF-2, FGF-7) and their receptors (FGFR-1IIIc, FGFR-2IIIb, FGFR-2IIIc), vascular endothelial growth factor (VEGF) and receptors (VEGFR-1, VEGFR-2), angiopoietin-tie-system (Angp-1, Angp-2, Tie-1, Tie-2) and nitric-oxid-synthase (eNOS, iNOS) were measured by qRT-PCR. An immunohistochemically determination and localisation was carried out for FGF-2, VEGF and VEGFR-1. Molecular size of FGF-2 was determined by westernblot. mRNA expression of the selected factors in pig total follicles and expression intensity in comparison to stromal tissue and CL was checked in experiment 1. Experiment 2 was concerned with the exact regulation of growth factors in the follicle compartments granulosa cells (GC) and theca interna (TI) during final follicular growth. A classification of follicles was performed according to follicular fluid oestradiol-17β (E2)-, progesterone and prostaglandin F2α content into four groups (2-3mm, 4-6mm, >7mm, post LH). Expression of FSHR decreased during follicle growth, while LHR and Aromatase increased. mRNA of FGF-family members was strongly expressed in stromal tissue. This was confirmed immunohistochemically by dominant localisation of FGF-2 protein in stromal cells. 18kDa and 22kDa isoforms of FGF-2 were overriding determined in stromal tissue. VEGF was located in GC and TI but not in stromal tissue. The receptor, VEGFR-1 was found in the endothelial- and smooth muscle cells of blood vessels. VEGF mRNA expression decreased after LH-peak. VEGFR-2 was upregulated in large follicles, VEGFR-1 after LH-peak. Angp-2/Angp-1 ratio was highest in small follicles, lowest in large follicles. After LH-peak the ratio rose again. Expression of eNOS was opposite with iNOS, which was downregulated in large follicles. FSH plays an important part in recruiting a folliclepool at time of luteolysis, while the further developing follicles become LH dependent. The number of LHR is an important selection criteria for the follicles. LH and E2 cause GC proliferation by mediators like FGF-7 and enhance angiogenesis by stimulating VEGF. While the follicle growth-up fast the pressure of oxygen in the follicular antrum decreases, whereby angiogenetic factors like VEGF and angiopoietins get stimulated and provide an adequate blood supply for the follicles. Activated by LH and VEGF eNOS is producing NO, which controls E2 synthesis and leads to a better blood flow in the TI through vasodilatation. Increasing E2 concentrations in large follicles stimulate Angp-1, which stabilise the now sufficient bloodvessel network in the TI. After the LH-peak Angp-2 destabilises the blood vessels, which is important for the process of ovulation and the succeeding angiogenesis in CL buildup. For follicle selection an important parameter seems to be the localisation of VEGF and FGF in the follicle. VEGF acts in the follicle and therefore follicles which express more VEGF and receptors have a developmental advantage. In cows this fact is suggested as one basic in selection of the one dominant follicle. FGF-2 in the pig is mainly expressed in stromal tissue and able to activate FGFR in the TI and increases angogenesis in a synergistic way with VEGF and its receptors. FGF-2 in stromal tissue consequently has influence on a few follicles and is maybe an important component in selection of follicles during final follicular growth in polyovulatory species.
